Endogenous and exogenous oxygenation of zwitterionic lipid membranes was studied by means of Fourier transform infrared (FTIR) spectroscopy. Spectra in the O-H stretching region were obtained for zwitterionic lipids (DLPC and DMPE) and for a dicationic lipid (DLPC-pant). In both the dicationic and zwitterionic systems, there was evidence for the formation of both long-lived and long-lived peroxyl radicals, the long-lived radicals being more prevalent in the zwitterionic lipids. The long-lived peroxyl radicals react with the membrane lipids to form the corresponding peroxides. The most reactive radicals are those with H atoms and are formed early in the reaction. The interaction of radicals and lipids was influenced by the lipid concentration, and there was little difference between endogenous and exogenous O2 in its effects on the radicals formed. The effect of the lipid concentration on the extent of lipid oxidation was also studied in the exogenous system. At lower lipid concentrations, peroxyl radicals were produced with small lipid hydroperoxide production. At higher lipid concentrations, lipid peroxidation occurred but with large amounts of hydroperoxide being
